Biological Background: RAB28 Function and Localization
- RAB28 is a member of the Ras-related Rab subfamily of small GTPases, which function as molecular switches cycling between GDP-bound inactive and GTP-bound active states to regulate intracellular membrane trafficking events, including vesicle formation, transport, tethering, and fusion. - Specifically, RAB28 has been implicated in the shedding and phagocytosis of cone cell outer segment discs in the retina (By similarity).
- Additionally, RAB28 participates in the nuclear transport of NF-kB p65/RELA in endothelial cells (By similarity).
- Subcellularly, RAB28 localizes to multiple compartments including the cell membrane, cytoplasm, cytoskeleton, cilium basal body, and nucleus.
- Three protein isoforms exist due to alternative splicing: isoform S is detected in most tissues; isoform L2 is widely and abundantly expressed; and isoform 3 is highly expressed in heart, lung, bone marrow, retina, brain, and retinal pigment epithelium.
- Post-translational modifications include prenylation, acetylation, and methylation; RAB28 is a GTP-binding protein linked to cone-rod dystrophy type 18 (CORD18).
