Biological Background: RAB26 Function and Localization
- RAB26 is a member of the Rab family of small GTPases, which are key regulators of intracellular membrane trafficking, controlling vesicle formation, movement, tethering, and fusion.
- Like other Rab proteins, RAB26 cycles between an inactive GDP-bound state and an active GTP-bound state, allowing it to recruit downstream effectors to membranes.
- RAB26 specifically mediates the transport of alpha-2A and alpha-2B adrenergic receptors (ADRA2A and ADRA2B) from the Golgi apparatus to the plasma membrane. Related references: PMID:23105096
- It plays an essential role in the maturation of zymogenic granules and in pepsinogen secretion within the stomach. Related references: PMID:20038531
- In the parotid gland, RAB26 is involved in the secretion of amylase from acinar granules (by similarity).
- At the subcellular level, RAB26 localizes to Golgi apparatus membranes and secretory vesicle membranes.
- The protein is predominantly expressed in brain tissue, indicating specialized functions in neuronal secretion.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–60) of human RAB26, a sequence conserved across human, mouse, and rat, supporting the reported cross-reactivity.
- In Western blot analysis under reducing conditions, a primary band near 28 kDa (predicted molecular weight) may be observed; additional bands could represent post-translationally modified forms or splice variants.
- For ELISA, consider pairing this antibody with an appropriate capture antibody or recombinant protein to establish a robust detection system; validate sensitivity and specificity in the relevant sample matrix.
- Given the predominant expression of RAB26 in brain, brain lysates or enriched membrane fractions may serve as suitable positive controls for WB.
