Biological Background: RAB24 Function and Localization

• The small GTPase Rab family, including RAB24, are key regulators of intracellular membrane trafficking, cycling between inactive GDP-bound and active GTP-bound forms. RAB24 is atypical, presenting low GTPase activity and existing predominantly in the GTP-bound active state.
• RAB24 is required for the clearance of late autophagic vacuoles under basal conditions, but is not needed for starvation-induced autophagy.
• RAB24 plays a role in meiotic apparatus assembly and progression during oocyte maturation, possibly via regulation of kinetochore-microtubule interactions.
• Subcellular localization includes cytoplasm, cytosol, membrane, autophagosome membrane, perinuclear region, and association with the spindle during meiosis.
• The protein contains a GTP-binding domain, is a phosphoprotein, undergoes prenylation, and requires magnesium as a cofactor.
