Biological Background: QKI Function and Localization
- QKI (protein quaking) is a KH domain-containing RNA-binding protein. It has multiple isoforms that localize to the nucleus, cytoplasm, and stress granules, enabling diverse regulatory roles.
- As an RNA reader, QKI binds specific RNAs to regulate pre-mRNA splicing, circular RNA formation, mRNA export, stability, and translation. It recognizes the 5'-NACUAAY-N(1,20)-UAAY-3' core sequence. Related references: PMID:22398723, PMID:23630077, PMID:25768908
- QKI plays a central role in myelinization by stabilizing myelin-related mRNAs (e.g., MBP, SIRT2), regulating mRNA transport, and indirectly controlling MAG alternative splicing. Related references: PMID:16641098
- QKI is involved in mRNA storage into stress granules, apoptosis, lipid deposition, interferon response, and glial cell development. Related references: PMID:25768908, PMID:31829086, PMID:34428287
- A cytosolic isoform recognizes N7-methylguanine (m7G) modified mRNAs; interaction with G3BP1 localizes these mRNAs to stress granules, suppressing translation. Related references: PMID:37379838
- QKI promotes monocyte differentiation via pre-mRNA splicing regulation, and delays macrophage differentiation by promoting CSF1R mRNA degradation. Related references: PMID:27029405, PMID:22398723
- QKI is expressed in the frontal cortex and is down-regulated in the brain of schizophrenic patients.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 242-341 of human QKI; consider this epitope when designing experiments or selecting detection modalities.
- For WB, a band around 38 kDa is expected for human QKI; validate in the relevant sample system and optimize loading controls.
- IHC-P and IF/ICC may require optimization of antigen retrieval methods and permeabilization conditions for your specific tissues or cells.
- The antibody cross-reacts with mouse and rat QKI, enabling multi-species studies; verify cross-reactivity in your model system as needed.
- For IP, consider using appropriate isotype controls and confirm specificity with downstream applications.
- ELISA can be employed for quantification; ensure the peptide epitope is accessible under your assay conditions.
