Biological Background: Pyruvate carboxylase (PC) Function and Localization
- Pyruvate carboxylase (PC) is a mitochondrial enzyme that catalyzes the ATP-dependent carboxylation of pyruvate to oxaloacetate, a critical anaplerotic reaction replenishing the tricarboxylic acid cycle intermediates.
- The reaction occurs in two steps: the biotin prosthetic group is first carboxylated using bicarbonate and ATP, followed by transfer of the carboxyl group to pyruvate to form oxaloacetate.
- PC plays a tissue-specific role in gluconeogenesis in the liver and kidney, and in de novo lipogenesis in adipose tissue, liver, and brain, linking carbohydrate and lipid metabolism.
- Located in the mitochondrial matrix, PC is synthesized as a precursor with a transit peptide that directs it to mitochondria.
- PC is a biotin-dependent, multifunctional ligase that requires manganese or magnesium as a metal cofactor and is regulated by acetylation and phosphorylation.
- Mutations in the PC gene are associated with pyruvate carboxylase deficiency, a rare autosomal recessive metabolic disorder.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 879–1178 of human PC, targeting the C-terminal region; consider evaluating reactivity with endogenous PC in the relevant species and tissue.
- For Western blotting, a band near 130 kDa is expected; include appropriate molecular weight markers and validate with positive controls.
- Optimize antibody dilution for each application (WB, IF/ICC, IP, ELISA) using a titration series and appropriate blocking conditions.
- Cross-reactivity is reported for human, mouse, and rat PC; due to polyclonal nature, lot-to-lot consistency should be verified in the intended assay.
