Biological Background: Peroxidasin Homolog (PXDN) Function and Localization
- Peroxidasin homolog (PXDN) is a secreted heme-containing peroxidase belonging to the peroxidase family, also known as melanoma-associated antigen MG50, vascular peroxidase 1, and p53-responsive gene 2 protein.
- PXDN catalyzes the two-electron oxidation of bromide by hydrogen peroxide to generate hypobromite, which mediates the formation of sulfilimine cross-links between methionine and hydroxylysine residues in collagen IV NC1 hexamers, essential for collagen IV network assembly. - These sulfilimine cross-links are critical for the collagen IV-dependent assembly of fibronectin and laminin, and for full extracellular matrix (ECM)-mediated signaling, including growth factor-induced cell proliferation and survival in endothelial cells, thus contributing to basement membrane integrity. - Subcellularly, PXDN localizes to the endoplasmic reticulum and is secreted into the extracellular space, incorporating into the extracellular matrix and basement membrane.
