Biological Background: PXDN Function and Localization
- PXDN (Peroxidasin homolog), also known as MG50, VPO1, and PRG2, is a secreted peroxidase belonging to the heme peroxidase family. An alternatively spliced isoform lacking the signal sequence is expressed in testis and in cells undergoing p53-dependent apoptosis, suggesting context-specific functional forms.
- Catalyzes bromide oxidation to hypobromite, which drives sulfilimine cross-link formation between methionine and hydroxylysine residues in collagen IV NC1 hexamers. This cross-linking is critical for collagen IV network assembly, fibronectin and laminin deposition, and extracellular matrix (ECM)-mediated signaling essential for basement membrane integrity.
