Biological Background: Nectin-3 Function and Localization
- Nectin-3 (also known as PVRL3, CD113, CDw113) is an immunoglobulin superfamily cell adhesion molecule that mediates cell-cell adhesion through heterophilic trans-interactions with other nectins and nectin-like proteins. Related references: PMID:16216929
- Trans-interaction with NECTIN2 at Sertoli-spermatid junctions and with PVR activates CDC42 and RAC small G proteins via SRC and RAP1 signaling, while also inducing endocytosis-mediated down-regulation of PVR from the cell surface, leading to reduced cell movement and proliferation. - Involved in axon guidance by promoting contacts between commissural axons and floor plate cells (By similarity).
- Contributes to the formation of cell-cell junctions, including adherens junctions and synapses (By similarity).
- Promotes a checkerboard-like cellular pattern in the auditory epithelium via heterophilic interaction with NECTIN1, supporting the organization of hair cells and supporting cells (By similarity).
- Predominantly expressed in testis, placenta, and many epithelial cell lines.
- Localized at the cell membrane, postsynaptic cell membrane, and adherens junctions; contains key structural features such as disulfide bonds, immunoglobulin domains, and a transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 127–366 of human PVRL3, a region within the extracellular domain; the resulting polyclonal antibody may recognize multiple splice variants and will likely target epitopes conserved in mouse.
- For Western blot, expect a band around 61 kDa in human and mouse samples; always validate specificity in your own experimental system.
- For ELISA, consider a sandwich or direct format and include appropriate negative and positive controls to confirm reactivity.
- Cross-reactivity with mouse is reported, but detectability in other species should be verified independently.
