Biological Background: PVRIG Function and Localization
- PVRIG is a transmembrane protein also known as CD112 receptor (CD112R) and poliovirus receptor-related immunoglobulin domain-containing protein.
- It functions as a cell surface receptor for NECTIN2, acting as a coinhibitory receptor that suppresses T-cell receptor-mediated signals, leading to inhibition of T-cell proliferation.
- The receptor competes with CD226 for binding to NECTIN2, modulating immune cell activation.
- PVRIG is localized at the cell membrane, consistent with its role in cell surface signaling.
- It is primarily expressed in immune cells, with detectable levels on CD8+ T-cells (mainly memory/effector) and NK cells, but not on B-cells, naive or helper T-cells, monocytes, neutrophils, or dendritic cells.
- Post-translational modifications include phosphorylation, as indicated by the keyword “Phosphoprotein”.
- The protein has a predicted molecular weight of approximately 34 kDa and contains a transmembrane helix.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 193-326) of human PVRIG, which may include the extracellular domain and can be suitable for detecting the receptor via Western blot (WB) or ELISA.
- For Western blot analysis of endogenous PVRIG, consider using lysates from CD8+ T-cell lines or NK cells, as these are reported to express the protein.
- Polyclonal antibodies may exhibit batch-to-batch variation; validate specificity in your assay system using appropriate controls.
- ELISA assay conditions should be optimized for antigen coating and antibody dilution.
