Biological Background: Pumilio 1 Function and Localization
- Sequence-specific RNA-binding protein that recognizes the Pumilio Response Element (PRE) 5'-UGUANAUA-3' in target mRNA 3'-UTRs. Related references: PMID:18328718, PMID:21397187, PMID:21572425
- Acts as a post-transcriptional repressor via two main mechanisms: direct recruitment of the CCR4-POP2-NOT deadenylase complex for mRNA degradation, and facilitation of miRNA binding to target sites (e.g., promoting miR-221/222 access to CDKN1B/p27 mRNA upon phosphorylation). Related references: PMID:22955276, PMID:18776931, PMID:20818387, PMID:20860814
- Represses key target mRNAs including E2F3 (involved in genomic stability) and ATXN1 (linked to neuronal function). Related references: E2F3: PMID:22345517, PMID:29474920; ATXN1: PMID:25768905
- Activity is negatively regulated by the long non-coding RNA NORAD, which sequesters PUM1 in the cytoplasm to prevent repression of mitotic, DNA repair, and replication factors. Related references: PMID:26724866
- Localizes predominantly in the cytoplasm, including P-bodies and cytoplasmic granules.
- Expressed in brain, heart, kidney, muscle, intestine, stomach, and all fetal tissues, but not in cerebellum, corpus callosum, caudate nucleus, hippocampus, medulla oblongata, or putamen.
- Additional functions: cytoplasmic sensing of viral infection and regulation of miRNA MIR199A biogenesis (in cooperation with PUM2). Related references: PMID:25340845, PMID:28431233
Experimental Guidance and Technical Tips
- For Western blot, the predicted molecular weight is 126 kDa; consider using a low-percentage gel (e.g., 6–8%) and extended transfer times to ensure efficient detection.
- In immunofluorescence/ICC, appropriate fixation (e.g., paraformaldehyde) and permeabilization (e.g., Triton X-100) may be required; validate subcellular localization in your specific cell model.
- The immunogen spans amino acids 104–203; this region is conserved between human and mouse, consistent with the reported cross-reactivity; verify reactivity in other species if needed.
- For ELISA applications, determine optimal antibody dilution by checkerboard titration; include appropriate positive and negative controls.
