Biological Background: PTRF Function and Localization

  • PTRF (Polymerase I and transcript release factor), also designated Cavin-1, CAVIN, FKSG13, or CGL4, is a caveolae-associated protein encoded by the CAVIN1 gene in humans.
  • Plays an essential role in caveolae formation and organization in all tissues. As a core component of the CAVIN complex, it is required for caveolae biogenesis in the presence of caveolin-1 (CAV1) and for proper CAV1 oligomerization. Related references: PMID:18056712; PMID:18191225; PMID:19726876
  • Regulates ribosomal transcription: enhances ribosomal transcriptional activity upon metabolic stimulation in adipocytes, contributes to ribosomal transcriptional loop formation, and releases paused RNA polymerase I and pre-rRNA from TTF1-bound DNA templates. Related references: PMID:18056712; PMID:18191225; PMID:19726876
  • Subcellular localization includes membrane, caveola, and cell membrane, as well as microsome, endoplasmic reticulum, cytoplasm, cytosol, mitochondrion, and nucleus.
  • Subject to multiple post-translational modifications: phosphorylation, acetylation, isopeptide bond formation, and ubiquitination.
  • Genetic variations in CAVIN1 are linked to congenital generalized lipodystrophy and diabetes mellitus, underscoring its importance in metabolic homeostasis.

Experimental Guidance and Technical Tips

  • The immunogen (residues 200–280 of human PTRF) targets a conserved central region, enabling potential cross-reactivity with mouse and rat orthologs.
  • For Western blot, anticipate a main band near 43 kDa; additional bands may arise from post-translational modifications or isoform diversity—validate using positive controls.
  • IHC-P and IF/ICC work best in caveolae-rich tissues (e.g., adipose, muscle, endothelium); consider nuclear/cytoplasmic staining patterns as the protein shuttles between compartments.
  • For ELISA applications, optimise antibody concentration and coating conditions using recombinant protein or cell lysates to assess linearity and specificity.
  • As a polyclonal preparation, lot-to-lot consistency should be verified, and antibody titration is recommended for each new assay.
