Biological Background: PTRF Function and Localization
- PTRF (Polymerase I and transcript release factor), also designated Cavin-1, CAVIN, FKSG13, or CGL4, is a caveolae-associated protein encoded by the CAVIN1 gene in humans.

- Regulates ribosomal transcription: enhances ribosomal transcriptional activity upon metabolic stimulation in adipocytes, contributes to ribosomal transcriptional loop formation, and releases paused RNA polymerase I and pre-rRNA from TTF1-bound DNA templates. - IHC-P and IF/ICC work best in caveolae-rich tissues (e.g., adipose, muscle, endothelium); consider nuclear/cytoplasmic staining patterns as the protein shuttles between compartments.
- For ELISA applications, optimise antibody concentration and coating conditions using recombinant protein or cell lysates to assess linearity and specificity.
