Biological Background: PTPRR Function and Localization

  • PTPRR (Receptor-type tyrosine-protein phosphatase R) is a member of the protein tyrosine phosphatase (PTP) family, also known by aliases including PTPRQ, EC-PTP, PCPTP1, PTP-SL, and PTPBR7.
  • Its primary function is to sequester mitogen-activated protein kinases (MAPKs) such as MAPK1, MAPK3, and MAPK14 in the cytoplasm, maintaining them in an inactive form; phosphorylation of the kinase interaction motif by PKA promotes MAPK release and nuclear translocation (By similarity).
  • The protein exhibits multiple subcellular localizations: it is found at the cell membrane, in the perinuclear cytoplasmic region, and is also secreted.
  • Post-translational modifications include phosphorylation and glycosylation; it contains an N-terminal signal peptide and a transmembrane domain, consistent with a receptor-type phosphatase topology (Keywords: Glycoprotein, Phosphoprotein, Signal, Transmembrane).
  • Tissue expression is broad: detected in brain, placenta, small intestine, stomach, uterus, and weakly in prostate; notably, the protein is present in cerebrospinal fluid (PubMed:25326458). Experimental Guidance and Technical Tips

  • For Western blot, the predicted molecular weight is 74 kDa; however, alternative splicing may produce isoforms of different sizes. Validate using appropriate positive controls and consider isoform-specific references.
  • The immunogen (aa22–227) lies within the N-terminal region shared by multiple isoforms; confirm specificity by knockdown or isoform-specific antibodies where necessary.
  • Validated for ELISA; when developing a sandwich immunoassay, screen the antibody in combination with a suitable detection antibody for human and rat samples.
  • High sequence homology between human and rat PTPRR supports observed cross-reactivity; always validate performance in the target species under your experimental conditions.
