Biological Background: PTPRN2 Function and Localization

  • PTPRN2 (Receptor-type tyrosine-protein phosphatase N2), also known as IA-2beta, phogrin, IAR, and ICAAR, is a type I transmembrane protein belonging to the protein-tyrosine phosphatase family.
  • It plays a crucial role in vesicle-mediated secretory processes, being required for normal accumulation of secretory vesicles in the hippocampus, pituitary, and pancreatic islets (By similarity).
  • Required for the accumulation of normal levels of insulin-containing vesicles and preventing their degradation, thus playing a role in insulin secretion in response to glucose stimuli (By similarity).
  • Involved in neurotransmitter accumulation in the brain, required for normal levels of norepinephrine, dopamine, and serotonin (By similarity).
  • Exhibits phosphatidylinositol phosphatase activity, dephosphorylating PI(4,5)P2, which regulates actin dynamics and cell migration (PubMed:26620550). Related references: PMID:26620550
  • Subcellularly localized to the membrane of secretory vesicles, including synaptic vesicles and secretory granule membranes.
  • Highest expression in brain and pancreas, with lower levels in trachea, prostate, stomach, and spinal cord (PubMed:8798755, PubMed:8954911). Related references: PMID:8798755, PMID:8954911
  • Key post-translational modifications include glycosylation, acetylation, and phosphorylation; alternative splicing produces multiple isoforms.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal extracellular region (aa 22–242), likely exposed in both denaturing and native conditions, making the antibody suitable for WB, IHC-P, and ELISA.
  • For Western blot, the full-length protein migrates at ~111 kDa; ensure adequate gel separation and transfer conditions.
  • For IHC-P, antigen retrieval may be considered and validation in positive control tissues such as pancreas or brain is recommended.
  • Cross-reactivity with mouse and rat enables comparative studies across species; however, verify specificity in the relevant experimental system.
