Biological Background: PTPIP51/RMDN3 Function and Localization

  • PTPIP51/RMDN3 (Regulator of microtubule dynamics protein 3), also known as Cerebral protein 10, Protein FAM82A2, Protein FAM82C, Protein tyrosine phosphatase-interacting protein 51, TCPTP-interacting protein 51, is encoded by the RMDN3 gene (formerly FAM82A2, FAM82C, PTPIP51). Alternative splicing may produce multiple isoforms.
  • Functionally, RMDN3 is involved in cellular calcium homeostasis regulation. It may participate in differentiation and apoptosis of keratinocytes, and its overexpression induces apoptosis.
  • Subcellularly, RMDN3 localizes to the mitochondrial outer membrane, cytoplasm, nucleus, and cytoskeleton including the spindle and spindle poles, suggesting roles in mitochondrial dynamics, calcium signaling, and cell division.
  • Tissue expression studies show high levels in epidermis and seminiferous epithelium: basal cells and spermatogonia show no perceptible amount, while suprabasal keratinocytes and differentiating spermatocytes up to spermatids exhibit high expression. Also present in fast twitch skeletal muscle fibers, ciliated epithelia associated with microtubular structures, and specific central nervous system regions (hippocampal neurons, autonomic ganglion cells, peripheral nerve axons). Widely expressed.
  • The protein contains coiled-coil domains and a transmembrane helix, consistent with its membrane association and potential protein-protein interactions. It is a phosphoprotein, with phosphorylation likely regulating its functions.
  • Keywords include: 3D-structure, Alternative splicing, Apoptosis, Coiled coil, Cytoplasm, Cytoskeleton, Differentiation, Membrane, Microtubule, Mitochondrion, Mitochondrion outer membrane, Nucleus, Phosphoprotein, Proteomics identification, Reference proteome, Transmembrane, Transmembrane helix.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 171-470, a region likely conserved across isoforms. For western blotting (WB), a band around 52 kDa is expected; consider using appropriate positive controls (e.g., HeLa, HEK293, or tissue lysates with high expression such as skin or testis).
  • For immunohistochemistry (IHC), expression is reported in stratified epithelia and testis; consider antigen retrieval optimization and validate staining specificity in these tissues.
  • For immunofluorescence (IF) or immunocytochemistry (ICC), note mitochondrial, nuclear, and cytoskeletal localization; co-staining with organelle markers may help confirm subcellular distribution.
  • ELISA performance should be validated with recombinant protein or cell lysates; consider testing serial dilutions to optimize signal-to-noise ratio.
