Biological Background: PTK2B Function and Localization
- Non-receptor tyrosine kinase that regulates reorganization of the actin cytoskeleton, cell polarization, migration, adhesion, spreading and bone remodeling.
- Plays a role in the regulation of the humoral immune response, required for normal marginal B-cell levels in spleen and migration of splenic B-cells, and contributes to macrophage polarization and T-cell responses.
- Functions in signaling downstream of integrin and collagen receptors, immune receptors, G protein-coupled receptors (GPCRs), and cytokine/chemokine/growth factor receptors, mediating responses to cellular stress.
- Forms multisubunit signaling complexes with SRC and SRC family kinases, leading to phosphorylation of numerous substrates and activation of pathways including PI3K/AKT1, NOS3, and MAPK cascades (MAPK1/ERK2, MAPK3/ERK1, MAPK8/JNK1).
- Recruits the ubiquitin ligase MDM2 to P53/TP53 in the nucleus, regulating P53 activity, ubiquitination and proteasomal degradation; also phosphorylates NMDA receptors and potassium channel subunits.
- Localizes to cytoplasm, perinuclear region, cell membrane, focal adhesions, lamellipodium, cell cortex and nucleus.
- Most abundant in the brain, with highest levels in amygdala and hippocampus; also expressed in spleen, lymphocytes, and at lower levels in kidney.
- Alternative splicing produces multiple isoforms; classified as a phosphoprotein and transferase.
