Biological Background: PTH1R Function and Localization

  • PTH1R (parathyroid hormone 1 receptor), also known as PTH/PTHrP type I receptor or PTH1 receptor, is encoded by the PTH1R gene and belongs to the class B G protein-coupled receptor family.
  • It functions as a receptor for parathyroid hormone (PTH) and parathyroid hormone-related peptide (PTHLH), playing critical roles in calcium homeostasis and bone remodeling. Related references: PMID:10913300 PMID:18375760 PMID:19674967
  • Upon ligand binding, PTH1R couples to G(s) alpha proteins to activate adenylate cyclase, leading to increased intracellular cAMP levels. Related references: PMID:20172855 PMID:30975883 PMID:35932760
  • PTHLH dissociates from the receptor more rapidly than PTH, resulting in a faster decay of the cAMP response, which may have implications for differential signaling dynamics. Related references: PMID:35932760
  • PTH1R is localized primarily at the cell membrane as a multi-pass transmembrane protein.
  • It is expressed in most tissues, with highest abundance in kidney, bone, and liver, consistent with its role in mineral ion homeostasis.
  • Post-translational modifications include glycosylation, phosphorylation, and disulfide bond formation, which may affect receptor stability and trafficking.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal extracellular domain (aa 27–188), which is exposed on the cell surface; the antibody may recognize both native and denatured forms under appropriate conditions.
  • For Western blotting, a band around 66 kDa is expected, but glycosylation may cause an upward shift; reducing conditions are typically recommended.
  • For immunohistochemistry (IHC-P), antigen retrieval and antibody titration are critical; frozen sections may also be tested for optimal signal.
  • In ELISA, the antibody can be used for direct or indirect detection; pairing with a detection antibody may require empirical optimization.
  • Cross-reactivity with mouse and rat has been reported, but sequence homology in the immunogen region should be verified for your specific experiment.
