Biological Background: PTGR3/ZADH2 Function and Localization
- PTGR3, also known as ZADH2 or PRG-3, is a member of the zinc-binding alcohol dehydrogenase family and functions as a prostaglandin reductase.
- It catalyzes the 13-reduction of 15-keto-prostaglandins, with the highest efficiency for 15-keto-PGE2-alpha, playing a critical role in prostanoid metabolism.
- Overexpression of PTGR3 inhibits adipocyte differentiation by repressing the transcriptional activity of PPARG.
- The protein localizes to peroxisomes, consistent with its involvement in lipid metabolism.
- PTGR3 is subject to post-translational modifications such as acetylation and phosphorylation, which may regulate its activity.
- The gene undergoes alternative splicing, leading to multiple protein isoforms.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 33–371 of human PTGR3 (NP_787103.1), encompassing nearly the full-length protein; this design should confer high specificity and sensitivity in detection assays.
- For Western blotting, a band at approximately 40 kDa is expected; consider using whole-cell lysates from human, mouse, or rat tissues/cell lines known to express PTGR3.
- In immunohistochemistry (IHC-P), optimize antigen retrieval and antibody dilution; validate in peroxisome-rich tissues such as liver or kidney.
- For ELISA, this antibody can be used as a capture or detection reagent; pair with a suitable monoclonal antibody if developing a sandwich assay for PTGR3 quantification.
- Since the antibody is a rabbit polyclonal, it may exhibit lot-to-lot variability; include appropriate positive and negative controls in each experiment.
