Biological Background: PTEN Function and Localization
- PTEN (phosphatase and tensin homolog) is a dual-specificity protein phosphatase that dephosphorylates tyrosine-, serine-, and threonine-phosphorylated proteins. Related references: PMID:9187108 PMID:9256433 PMID:9616126

- The immunogen corresponds to the C-terminal region (residues 304–403) of human PTEN; conservation across mouse and rat makes it likely suitable for multi-species detection in Western blot and IHC applications.
- In Western blot (WB), PTEN may produce multiple bands (~20 kDa, 47 kDa, and 65 kDa) due to isoforms and post-translational modifications; consider including phosphatase inhibitors in lysis buffer to preserve phosphorylation states.
- For immunohistochemistry (IHC-P), antigen retrieval conditions (e.g., citrate buffer pH 6.0 or EDTA pH 8.0) should be optimized empirically, and cross-reactivity with rodent tissues validated on relevant controls.
- In immunoprecipitation (IP), the rabbit IgG origin allows compatibility with standard Protein A/G beads; confirm the IgG subclass for optimal bead selection.
- For immunofluorescence/immunocytochemistry (IF/ICC), permeabilization with 0.1–0.5% Triton X-100 and blocking with 5% BSA may help reduce non-specific background.
