Biological Background: PSRC1 Function and Localization
- PSRC1 (Proline/serine-rich coiled-coil protein 1), also known as DDA3 or FP3214, is a mitotic spindle-associated protein.
- It is required for normal progression through mitosis, facilitating proper chromosome congression at the metaphase plate and ensuring normal rates of chromosomal segregation during anaphase.
- PSRC1 regulates mitotic spindle dynamics by increasing the turnover of microtubules on metaphase spindles, contributing to the generation of normal tension across sister kinetochores.
- It recruits the microtubule depolymerase KIF2A and the spindle pole protein ANKRD53 to the mitotic spindle and spindle poles, thereby modulating spindle assembly and stability.
- Subcellularly, PSRC1 localizes to the cytoplasm, the mitotic spindle, and spindle poles during cell division.
- Widely expressed in adult and fetal tissues, with highest levels in the adult brain and fetal thymus; it is not detected in adult skeletal muscle.
- PSRC1 may also participate in p53/TP53-regulated growth suppression, linking cell cycle control to tumor suppression pathways.
