Biological Background: PSME3 Function and Localization

  • PSME3, also known as PA28gamma, REG-gamma, or Ki nuclear autoantigen, is the gamma subunit of the 11S proteasome activator complex.
  • It forms a doughnut-shaped homoheptamer that binds the 20S proteasome, enhancing trypsin-like activity while inhibiting chymotrypsin-like and postglutamyl-preferring activities.
  • PSME3 plays a critical role in regulating apoptosis by facilitating MDM2-mediated ubiquitination and proteasomal degradation of p53/TP53, limiting p53 accumulation after DNA damage.
  • It is also involved in cell cycle regulation and mediates CCAR2- and CHEK2-dependent inhibition of SIRT1. Related references: PMID:25361978
  • Subcellularly, PSME3 localizes to both the nucleus and the cytoplasm.
  • Post-translational modifications include acetylation and phosphorylation, and it is associated with host-virus interactions.

Experimental Guidance and Technical Tips

  • The immunogen used to generate this antibody corresponds to a C-terminal peptide (aa 200–254) of human PSME3. Verify sequence homology if cross-reactivity with other species or isoforms is critical.
  • For Western blot (WB), endogenous PSME3 migrates at approximately 30 kDa. Use appropriate loading controls and validate antibody specificity in your sample system.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and permeabilization conditions. Include appropriate negative controls to confirm staining specificity.
