Biological Background: PSMB7 Function and Localization
- PSMB7 (proteasome subunit beta type-7), also known as macropain chain Z or multicatalytic endopeptidase complex chain Z, is a core component of the 20S proteasome complex (UniProt Q99436).
- It participates in ATP-dependent degradation of ubiquitinated proteins within the 26S proteasome, maintaining protein homeostasis by removing misfolded, damaged, or temporally unneeded proteins.
- The subunit mediates ubiquitin-independent proteolysis when the 20S core associates with regulatory particles like PA200 or PA28, important for processes such as spermatogenesis and MHC class I antigen presentation.
- PSMB7 exhibits trypsin-like catalytic activity, cleaving peptide bonds after basic residues.
- The protein localizes to both the cytoplasm and nucleus.
- Expression is low in normal colonic mucosa but significantly upregulated in colorectal cancer tissues, implicating PSMB7 in tumorigenesis.
- The protein is synthesized as a zymogen precursor that undergoes autocatalytic processing to generate the mature active subunit.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fragment covering amino acids 44–200, which includes part of the mature subunit; the antibody may recognize both precursor (≈30 kDa) and mature forms.
- For Western blot, a single band near 30 kDa is expected; consider validating specificity using siRNA knockdown or knockout cell lysates.
- For immunofluorescence/ICC, optimize fixation and permeabilization conditions; nuclear and cytoplasmic staining patterns may be observed.
- In ELISA applications, verify the antibody's performance as a capture or detection reagent through standard pairing experiments.
- Cross-reactivity with mouse and rat PSMB7 allows multi-species studies; however, always confirm reactivity in your experimental system.
