Biological Background: PSMA/FOLH1 Function and Localization

  • PSMA/FOLH1, also known as glutamate carboxypeptidase 2, folate hydrolase 1, and NAALADase I, is a multifunctional zinc metalloprotease encoded by the FOLH1 gene. It exhibits both folate hydrolase and N-acetylated-alpha-linked-acidic dipeptidase (NAALADase) activity.
  • The enzyme preferentially hydrolyzes tri-alpha-glutamate peptides. In the intestine, it is essential for dietary folate uptake; in the brain, it modulates excitatory neurotransmission by hydrolyzing the neuropeptide NAAG to release glutamate.
  • PSMA/FOLH1 also possesses dipeptidyl-peptidase IV type activity, as it cleaves Gly-Pro-AMC in vitro.
  • The protein is a single-pass type II membrane protein localized predominantly at the cell membrane, but it is also found in the cytoplasm.
  • FOLH1 is highly expressed in prostate epithelium and detected in a wide range of tissues including urinary bladder, kidney, testis, ovary, fallopian tube, breast, adrenal gland, liver, esophagus, stomach, small intestine, colon, and brain. It is also expressed in the capillary endothelium of various tumors.
  • In prostate cancer, distinct isoforms are expressed: isoform PSMA-1 is most abundant in primary tumors, and isoform PSMA-9 is specifically expressed in prostate cancer, while isoform PSMA' predominates in normal prostate.
  • The protein undergoes post-translational modifications including glycosylation and phosphorylation, and requires zinc and calcium ions for its catalytic activity.

Experimental Guidance and Technical Tips

  • The immunogen is a recombinant fusion protein encompassing the N-terminal cytoplasmic domain (amino acids 1–106) of human PSMA/FOLH1. This region is relatively conserved; however, cross-reactivity with rat is reported, so validation in the relevant species is recommended.
  • For Western blot, a band around 84 kDa is expected. Given the glycoprotein nature, the observed band may appear slightly larger or broader. Optimal blocking and antibody dilution should be determined empirically.
  • In immunofluorescence (IF-P), consider using permeabilization to allow access to the intracellular epitope. Co-staining with a plasma membrane marker can help confirm localization.
  • For ELISA, the antibody may be used to capture or detect human PSMA/FOLH1 in samples such as cell lysates or tissue extracts, but sandwich pair compatibility must be verified.
