Biological Background: PSMA/FOLH1 Function and Localization

  • PSMA/FOLH1 (Glutamate carboxypeptidase 2) is a multifunctional enzyme that possesses both folate hydrolase and N-acetylated-alpha-linked acidic dipeptidase (NAALADase) activity, with preference for tri-alpha-glutamate peptides.
  • In the intestine, it facilitates folate absorption by cleaving polyglutamated folates, while in the brain, it modulates glutamatergic neurotransmission by hydrolyzing the neuropeptide NAAG to release glutamate.
  • The protein is involved in prostate tumor progression, and isoform-specific expression has been observed: PSMA' predominates in normal prostate, PSMA-1 in primary tumors, and PSMA-9 is specifically expressed in prostate cancer.
  • Subcellularly, it is anchored to the cell membrane via a signal-anchor transmembrane domain, but also localizes to the cytoplasm.
  • It additionally exhibits dipeptidyl-peptidase IV-like activity, cleaving substrates such as Gly-Pro-AMC in vitro.
