Biological Background: PSMA5 Function and Localization
- PSMA5 (proteasome subunit alpha type-5), also referred to as macropain zeta chain, multicatalytic endopeptidase complex zeta chain, or proteasome zeta chain, is a core subunit of the 20S proteasome complex.
- As part of the 20S core proteasome, PSMA5 is involved in the proteolytic degradation of most intracellular proteins. When associated with two 19S regulatory particles to form the 26S proteasome, it facilitates ATP-dependent degradation of ubiquitinated proteins, crucial for protein homeostasis.
- In its PA200- or PA28-associated form, PSMA5 participates in ubiquitin-independent protein degradation, which is important for processes such as spermatogenesis and the generation of MHC class I-presented antigenic peptides.
- The PSMA5 gene is a member of the proteasome alpha subunit family and encodes a protein that localizes to both the cytoplasm and nucleus.
- PSMA5 is subject to several post-translational modifications, including acetylation, phosphorylation, and glycosylation, which may regulate its function.
- At the protein level, PSMA5 expression has been detected in fetal brain.
