Biological Background: PSMA1 Function and Localization
- PSMA1 encodes the proteasome subunit alpha type-1, also known as PROS-30, macropain subunit C2, or proteasome component C2. It is a core component of the 20S proteasome complex.
- As part of the 20S proteasome, PSMA1 is responsible for the proteolytic degradation of most intracellular proteins. The 20S core can associate with regulatory particles to form the 26S proteasome, which degrades ubiquitinated proteins in an ATP-dependent manner, maintaining protein homeostasis.
- The 20S proteasome also mediates ubiquitin-independent protein degradation when associated with PA200 or PA28 regulators, playing critical roles in spermatogenesis (20S-PA200) and MHC class I antigen presentation (20S-PA28).
- PSMA1 is localized in both the cytoplasm and nucleus, consistent with its broad role in protein turnover.
- The protein is subject to various post-translational modifications including acetylation, phosphorylation, and glycation, as well as ubiquitin conjugation.
- PSMA1 is involved in immunity through its role in generating antigenic peptides for MHC class I presentation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to full-length human PSMA1 (amino acids 1–263), which may provide broad epitope recognition. Consider validating the antibody using positive controls such as HeLa or HEK293 cell lysates.
- For Western blotting, start with a 1:1000 dilution and optimize as needed. The predicted molecular weight is approximately 30 kDa.
- When performing IF/ICC, note that PSMA1 localizes to both the cytoplasm and nucleus; include appropriate fixation and permeabilization steps for nuclear antigens.
- As the antibody cross-reacts with mouse and rat, validate its reactivity in your specific model system to confirm suitability.
- For ELISA applications, standard direct or indirect protocols can be considered; optimize coating antigen and antibody concentration.
