  • Postsynaptic scaffolding protein (PSD-95) encoded by DLG4, also known as synapse-associated protein 90 (SAP-90), plays a pivotal role in organizing synaptic protein complexes.
  • Critical for synaptogenesis and synaptic plasticity by clustering NMDA receptor subunits, shaker-type potassium channels, and other signaling molecules at the postsynaptic density.
  • Regulates AMPA-type glutamate receptor (AMPAR) immobilization, maintaining channels in an activated state in the presence of glutamate and preventing synaptic depression (By similarity).
  • Overexpression or depletion of DLG4 alters the ratio of excitatory to inhibitory synapses in hippocampal neurons, underscoring its role in balancing synaptic transmission.
