Biological Background: PSD95 Function and Localization
- Postsynaptic scaffolding protein (PSD-95) encoded by DLG4, also known as synapse-associated protein 90 (SAP-90), plays a pivotal role in organizing synaptic protein complexes.
- Critical for synaptogenesis and synaptic plasticity by clustering NMDA receptor subunits, shaker-type potassium channels, and other signaling molecules at the postsynaptic density.
- Regulates AMPA-type glutamate receptor (AMPAR) immobilization, maintaining channels in an activated state in the presence of glutamate and preventing synaptic depression (By similarity).
- Overexpression or depletion of DLG4 alters the ratio of excitatory to inhibitory synapses in hippocampal neurons, underscoring its role in balancing synaptic transmission.
- Cooperates with FYN to stabilize palmitoyltransferase ZDHHC5 at the synaptic membrane through FYN-mediated phosphorylation, inhibiting ZDHHC5 association with endocytic proteins. Related references: PMID:26334723

- In IHC-P, expect punctate synaptic staining patterns; optimize antigen retrieval and antibody dilution for your tissue type.
- Consider testing dilutions starting at 1:1000 for WB and 1:100 for IHC-P; validate performance with appropriate controls.
