Biological Background: PSD95 Function and Localization
- PSD95, encoded by the DLG4 gene, is a postsynaptic scaffolding protein essential for synaptogenesis and synaptic plasticity, interacting with NMDA receptors, potassium channels, and AMPA receptors.
- PSD95 is a member of the membrane-associated guanylate kinase (MAGUK) family, containing three PDZ domains, an SH3 domain, and a guanylate kinase-like domain, enabling multi-protein complex assembly.
- It localizes primarily to the postsynaptic density of excitatory synapses in the brain, with expression also detected in dendritic spines, axons, and presynaptic regions.
- PSD95 regulates AMPA receptor immobilization at the postsynaptic density, maintaining channels in an activated state to prevent synaptic depression (By similarity).
- It cooperates with FYN kinase to stabilize palmitoyltransferase ZDHHC5 at the synaptic membrane, preventing its endocytosis and thus ensuring proper palmitoylation of synaptic proteins. Related references: PMID:26334723

Experimental Guidance and Technical Tips
- The immunogen corresponds to the SH3 domain region (aa 420–510); therefore, the antibody may recognize all isoforms containing this region.
- For Western blot, a primary band at approximately 80 kDa is expected in brain tissue lysates; additional bands may represent isoforms or degradation products.
- For immunofluorescence, optimal fixation and permeabilization conditions should be empirically determined to visualize synaptic localization.
- This polyclonal antibody detects human, mouse, and rat PSD95, making it suitable for cross-species studies; however, expression levels vary across tissues, so include appropriate controls.
