Biological Background: PSA/KLK3 Function and Localization
- Prostate-specific antigen (PSA), also referred to as gamma-seminoprotein, kallikrein-3 (KLK3), P-30 antigen, or semenogelase, is a member of the tissue kallikrein family of serine proteases encoded by the KLK3 gene (GeneID 354).
- The protein is synthesized as a preproenzyme, processed into a zymogen, and ultimately activated by cleavage to yield the mature ~29 kDa glycoprotein (UniProt: P07288).
- PSA is predominantly expressed in prostate epithelial cells and secreted into seminal plasma and serum; its secreted nature is confirmed by the "Secreted" keyword and subcellular localization annotation.
- Its primary physiological function is the hydrolysis of semenogelin-1, the major gel-forming protein in semen, resulting in liquefaction of the seminal coagulum and facilitation of sperm motility.
- PSA belongs to the extended kallikrein family clustered on chromosome 19; alternative splicing of the KLK3 gene generates multiple isoforms.
- The protein is extensively post-translationally modified, including disulfide bond formation, glycosylation, and proteolytic processing from the zymogen; it contains a signal peptide for secretion.
- Structural studies (3D-structure keyword) have elucidated the catalytic triad and substrate-binding pocket typical of trypsin-like serine proteases.
- PSA has been directly identified by protein sequencing and proteomics, serving as a reference protein for the human proteome (keywords: Direct protein sequencing, Proteomics identification, Reference proteome).
Experimental Guidance and Technical Tips
- The immunogen corresponds to a sequence within the mature PSA protein (amino acids 162–261), avoiding the signal peptide and propeptide regions. This region is likely to generate antibodies recognizing both free PSA and PSA complexed to inhibitors.
- For Western blot (WB) analysis, a reducing SDS-PAGE protocol is recommended; expect a band corresponding to the mature protein at approximately 29 kDa. Prostate-derived cell lines (e.g., LNCaP) or human seminal plasma may serve as positive controls.
- When developing ELISA assays, pairing this monoclonal antibody with a detection antibody against a non-overlapping epitope may enhance sensitivity. Validate performance with recombinant PSA standards and clinical samples to confirm specificity.
- Note that this antibody is validated for human PSA; cross-reactivity with other kallikreins has not been reported.
