Biological Background: Peripherin (PRPH) Function and Localization
- Peripherin is a class-III neuronal intermediate filament protein, also known as Neurofilament 4 (NEF4).
- It forms an independent structural network or cooperates with other neurofilament proteins (NEFL, NEFH, NEFM, INA) to form filamentous networks; assembly may be regulated by RAB7A. Related references: PMID:15322088 PMID:15446584
- PRPH plays a role in the development of unmyelinated sensory neurons and may be involved in axon elongation and regeneration after injury.
- It inhibits neurite extension in type II spiral ganglion neurons in the cochlea.
- Subcellularly, peripherin localizes to the cytoplasm, cytoskeleton, axon, and perikaryon.
- Expressed predominantly in neurons of the outer hair cells in the organ of Corti and to a lesser extent in type I spiral ganglion cells.
- Post-translational modifications include phosphorylation and nitration; implicated in amyotrophic lateral sclerosis and neurodegeneration.

- For Western blot, the predicted molecular weight is approximately 54 kDa. Validate antibody specificity using PRPH-positive neuronal cell lines or tissue lysates.
