Biological Background: PRPF8 Function and Localization
- PRPF8 (Pre-mRNA-processing-splicing factor 8) is also known as PRP8 homolog, 220 kDa U5 snRNP-specific protein, and p220, encoded by the PRPF8 gene in humans.
- It functions as a core component of the precatalytic, catalytic, and postcatalytic spliceosomal complexes in both the major U2-type and minor U12-type spliceosomes, playing a critical role in pre-mRNA splicing. Related references: PMID:10411133 PMID:11971955 PMID:28076346
- Acts as a scaffold that mediates the ordered assembly of spliceosomal proteins and snRNAs, and is required for the formation of the U4/U6-U5 tri-snRNP complex.
- Positions spliceosomal U2, U5, and U6 snRNAs at splice sites on pre-mRNA substrates and interacts with both the 5' and 3' splice sites to facilitate splicing.
- Localizes to the nucleus, particularly in nuclear speckles, which are sites of pre-mRNA processing.
- Widely expressed across tissues, indicating its fundamental role in gene expression.
- Subject to post-translational modifications including acetylation, methylation, and phosphorylation, which may regulate its activity.
- Associated with retinitis pigmentosa, a disease variant, underscoring its clinical significance.
Experimental Guidance and Technical Tips
- For western blot (WB), note that PRPF8 has a predicted molecular weight of 274 kDa; use a low-percentage SDS-PAGE gel (e.g., 6–8%) and optimize transfer conditions for high-molecular-weight proteins.
- The immunogen covers amino acids 2200–2335, suggesting recognition of a C-terminal epitope. Consider this if using truncated constructs.
- Cross-reactivity is shown with human, mouse, and rat; validate in your specific sample system.
- Applications listed include WB, IHC-P, IF/ICC, and ELISA; optimize dilutions and conditions for each assay.
