Biological Background: PRP19 Function and Localization
- PRP19 (Pre-mRNA-processing factor 19), also known as Nuclear matrix protein 200, is a ubiquitin-protein ligase and core component of the spliceosome and DNA repair complexes.
- It is a RING-type E3 ubiquitin transferase that catalyzes Lys-63-linked polyubiquitination of spliceosomal protein PRPF3 to stabilize the U4/U5/U6 tri-snRNP complex. Related references: PMID:20595234
- PRP19 is part of the PRP19C/Prp19 complex/NTC/Nineteen complex required for spliceosome assembly and activity. Related references: PMID:28076346 PMID:28502770 PMID:29301961
- It participates in DNA damage response by ubiquitinating RPA1 and RPA2, facilitating ATR-ATRIP recruitment and ATR activation. Related references: PMID:24332808
- PRP19 localizes to the nucleus, nucleoplasm, and can be found in the cytoplasm, cytoskeleton (spindle), and lipid droplets.
- Tissue expression is ubiquitous, but it is weakly expressed in senescent cells and highly expressed in tumor cell lines, consistent with a role in senescence evasion.

- Cross-reactivity with mouse and rat is reported; verify expression levels in target tissues before proceeding.
