Biological Background: PROX1 Function and Localization

  • PROX1, or Prospero homeobox protein 1, is a transcription factor with a DNA-binding homeobox domain; it functions as a key regulator of developmental processes including cell fate determination, gene transcriptional regulation, and progenitor cell control in various organs.
  • It plays critical roles in embryonic development, particularly in neurogenesis and the formation of the heart, eye lens, liver, pancreas, and lymphatic system.
  • PROX1 is involved in circadian rhythm regulation by repressing transcription of the retinoid-related orphan receptor RORG, as well as the transcriptional activity of RORA and RORG, and by suppressing expression of core clock genes such as BMAL1, NPAS2, and CRY1, along with metabolic genes AVPR1A and ELOVL3.
  • The protein localizes predominantly to the nucleus, consistent with its role as a transcription factor.
  • PROX1 is most actively expressed in the developing lens, with additional detection in embryonic brain, lung, liver, and kidney; in adults, higher levels are found in heart and liver compared to brain, skeletal muscle, kidney, and pancreas.
  • Post-translational modifications include phosphorylation and ubiquitination (Ubl conjugation), and it is known to form isopeptide bonds, which may influence its stability and activity.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 220-375 of human PROX1, a region covering part of the homeobox domain that is conserved across species; cross-reactivity has been observed in human, mouse, and rat, but validate in your specific model.
  • For Western blotting, PROX1 has a predicted molecular weight of approximately 83 kDa; use appropriate lysis buffers and loading controls, and consider using embryonic tissue or cell lines known to express PROX1 for positive controls.
  • For ELISA applications, optimize coating antigen concentration and blocking conditions; include known positive and negative samples to establish assay specificity.
  • Given PROX1’s nuclear localization, ensure protein extraction protocols are compatible with nuclear protein recovery for accurate detection.
