Biological Background: Proteasome 20S LMP7/PSMB8 Function and Localization
- PSMB8 encodes the proteasome subunit beta type-8, also known as low molecular mass protein 7 (LMP7), which is a component of the immunoproteasome.
- This subunit replaces the constitutive PSMB5 in the immunoproteasome, enhancing the capacity to cleave peptides after hydrophobic and basic residues, and plays a critical role in generating antigenic peptides for MHC class I presentation. Involved in the generation of spliced peptides. Related references: PMID:27049119
- PSMB8 is a key mediator of interferon gamma-induced sensitivity and participates in apoptosis by degrading the anti-apoptotic protein MCL1.
- It is required for the differentiation of preadipocytes into adipocytes, indicating non-immune functions.
- The protein localizes to both the cytoplasm and nucleus.
- Post-translational modifications include phosphorylation, and it functions as a threonine protease that is initially synthesized as a zymogen.
- Alternative splicing produces isoforms (E1 and E2) with distinct functions; in cancer cells, substitution of isoform 1 by isoform 2 results in immunoproteasome deficiency.
Experimental Guidance and Technical Tips
- For Western blot (WB), the predicted molecular weight of PSMB8 is approximately 30 kDa; consider using a positive control lysate from cell lines or tissues known to express the immunoproteasome, such as IFN-γ-treated cells.
- In immunohistochemistry (IHC-P), antigen retrieval may be necessary to expose the epitope; optimize antibody dilution using a tissue known to express PSMB8, such as immune organs.
- For ELISA applications, the recombinant protein immunogen suggests suitability for both direct and sandwich formats; titrate the antibody to determine optimal working concentration.
- As a rabbit monoclonal antibody, expect high specificity and reproducibility; consider cross-adsorption if non-specific binding is observed in multispecies samples.
- Cross-reactivity with mouse and rat PSMB8 indicates potential for use in rodent models; however, confirm specificity in the target species' tissues.
