Biological Background: Prostatic Acid Phosphatase Function and Localization
- Prostatic acid phosphatase (PSAP), also known as ACPP, 5'-NT, or ACP-3, is a non-specific tyrosine phosphatase encoded by the ACP3 gene in humans.
- Catalyzes dephosphorylation of various substrates under acidic conditions (pH 4–6), including alkyl, aryl, and acyl phosphate monoesters, and phosphorylated proteins. Related references: PMID:10506173, PMID:15280042, PMID:20498373
- Acts as a lipid phosphatase, inactivating lysophosphatidic acid in seminal plasma. Related references: PMID:10506173, PMID:15280042, PMID:20498373
- Functions as a tumor suppressor in prostate cancer by dephosphorylating ERBB2 and suppressing MAPK-mediated signaling. Related references: PMID:20498373
- Has ecto-5'-nucleotidase activity in dorsal root ganglion neurons, generating adenosine from AMP to modulate pain.
- In seminal plasma, PSAP forms amyloid beta-sheet fibrils (SEVI) that capture and enhance HIV infection by attaching virions to target cells. Related references: PMID:18083097, PMID:19451623, PMID:19897482
- Subcellularly localized to the cell membrane, lysosome membrane, secreted fraction, nucleus, and cytoplasm.
- Highly expressed in prostate epithelial cells; also detected in bladder, kidney, pancreas, lung, cervix, testis, ovary, and skeletal muscle sarcolemma.
- Post-translationally modified by glycosylation and stabilized by disulfide bonds; classified as a glycoprotein with a signal peptide and transmembrane region.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 33–386 of human PSAP, covering the majority of the mature protein; this suggests broad epitope coverage, but monoclonal specificity should be confirmed in your assay system.
- For Western blotting, validate detection of the ~45 kDa band and note that glycosylation may cause a slightly higher apparent molecular weight; consider using deglycosylation controls if needed.
- For immunohistochemistry (IHC-P), optimize antigen retrieval conditions (e.g., heat-induced epitope retrieval in citrate or Tris-EDTA buffer) and test antibody dilution series on positive (prostate tissue) and negative controls.
- In ELISA, the antibody’s reactivity with human and rat PSAP may enable cross-species quantitative analysis; validate standard curves and assay specificity.
- Since this rabbit monoclonal antibody has been tested in WB, IHC-P, and ELISA, additional applications (e.g., IF, flow cytometry) should be piloted with appropriate fixatives and permeabilization methods.
- Store at –20°C upon receipt and avoid repeated freeze-thaw cycles; the product is shipped on ice bags to maintain stability.
