Biological Background: Prostatic Acid Phosphatase (ACPP) Function and Localization
- Prostatic acid phosphatase (ACPP), encoded by the ACP3 gene, is a secreted and membrane-associated protein also known as 5′-nucleotidase (5′-NT), acid phosphatase 3, and thiamine monophosphatase (TMPase). Alternative splicing produces multiple isoforms.
- It functions as a non-specific tyrosine phosphatase, dephosphorylating diverse substrates under acidic conditions (pH 4–6) and displaying lipid phosphatase activity that inactivates lysophosphatidic acid in seminal plasma. Related references: PMID:10506173, PMID:15280042, PMID:20498373
- ACPP acts as a tumor suppressor in prostate cancer by dephosphorylating ERBB2 and deactivating MAPK-mediated signaling pathways. Related references: PMID:20498373
- In dorsal root ganglion neurons, it exhibits ecto-5′-nucleotidase activity, generating the pain-suppressing nucleoside adenosine from AMP.
- In semen, ACPP can form SEVI amyloid fibrils that capture HIV virions and enhance infection by promoting viral attachment to target cells. - Subcellular localization includes secretion, as well as association with the cell membrane, lysosome membrane, nucleus, and cytoplasm/cytosol.
- Tissue expression is highest in prostate glandular and ductal epithelial cells, with additional expression in bladder, kidney, pancreas, lung, cervix, testis, ovary, and other tissues; it is widely expressed and found in the sarcolemma of skeletal muscle.

- The immunogen comprises amino acids 33–386 of the mature human ACPP protein, suggesting broad epitope coverage. This recombinant rabbit PolymAb may recognize both native and denatured forms, suitable for WB, IHC-P, and ELISA.
- For western blot, anticipate a band near 45 kDa (the predicted molecular weight of the precursor) or the processed mature form; consider that glycosylation may shift the apparent molecular weight.
- In IHC-P on formalin-fixed paraffin-embedded tissues, validate with appropriate positive (e.g., prostate) and negative controls, and consider standard antigen retrieval protocols.
- The antibody detects ACPP from human, mouse, and rat, enabling cross-species studies; however, verify expression levels in specific rodent tissues and cell lines.
