Biological Background: Prostatic Acid Phosphatase (ACPP) Function and Localization

  • Prostatic acid phosphatase (ACPP) is a non-specific tyrosine phosphatase also known as 5'-nucleotidase (5'-NT), acid phosphatase 3, and ecto-5'-nucleotidase, encoded by the ACP3 gene with multiple isoforms due to alternative splicing.
  • Functions as a phosphatase active at acidic pH (4-6), dephosphorylating alkyl, aryl, and acyl orthophosphate monoesters and phosphorylated proteins; also exhibits lipid phosphatase activity, inactivating lysophosphatidic acid in seminal plasma. Related references: PMID:10506173 PMID:15280042 PMID:20498373
  • Acts as a tumor suppressor in prostate cancer by dephosphorylating ERBB2 and deactivating MAPK-mediated signaling; additionally functions as an ecto-5'-nucleotidase in dorsal root ganglion neurons, generating adenosine from AMP for pain suppression. Related references: PMID:20498373
  • Forms amyloid beta-sheet fibrils (SEVI) in semen that capture HIV virions and enhance infection; SEVI assembly and HIV enhancement can be inhibited by EGCG, WW61 peptide, and surfen; also enhances XMRV infection. Related references: PMID:18083097 PMID:19451623 PMID:19897482 PMID:21677644 PMID:19403677
  • Subcellularly localizes to the cell membrane, lysosome membrane, nucleus, cytosol, and is also secreted.
  • Highly expressed in normal prostate glandular and ductal epithelial cells, with lower levels in prostate carcinoma; also detected in bladder, kidney, pancreas, lung, cervix, testis, ovary, and skeletal muscle.
  • Post-translationally modified as a glycoprotein with disulfide bonds; multiple isoforms exist due to alternative splicing and proteolytic processing.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 33–386 of human ACPP, which excludes the signal peptide; the antibody may recognize both full-length and processed forms.
  • For western blot (WB) with predicted protein weight of 45 kDa, consider using reducing conditions and validate in cell or tissue lysates with known expression.
  • For immunohistochemistry (IHC-P), antigen retrieval protocols may be necessary; testing on prostate tissue sections is recommended given high expression in normal prostate.
  • Given cross-reactivity with mouse and rat, validate antibody performance in these species by checking sequence homology and using appropriate controls.
  • As a polyclonal antibody, it may detect multiple isoforms or processing variants; consider confirming band specificity with siRNA or blocking peptides.
  • For ELISA applications, optimize coating concentrations and secondary detection systems; recombinant ACPP protein can serve as a positive control.
