Biological Background: Prostatic Acid Phosphatase (ACPP) Function and Localization

  • Prostatic acid phosphatase (ACPP), also designated as 5'-nucleotidase (5'-NT), acid phosphatase 3, ecto-5'-nucleotidase, and protein tyrosine phosphatase ACP3, is encoded by the ACP3 gene in humans.
  • Functions as a non-specific tyrosine phosphatase under acidic conditions (pH 4-6), hydrolyzing alkyl, aryl, and acyl orthophosphate monoesters as well as phosphorylated proteins; also possesses lipid phosphatase activity and inactivates lysophosphatidic acid in seminal plasma. Related references: PMID:10506173, PMID:15280042
  • Acts as a tumor suppressor in prostate cancer by dephosphorylating ERBB2 and deactivating MAPK-mediated signaling pathways. Related references: PMID:20498373
  • Exhibits ecto-5'-nucleotidase activity in dorsal root ganglion neurons, generating adenosine from AMP, which functions as a pain suppressor.
  • In the context of microbial infection, ACPP forms SEVI amyloid fibrils in semen that capture HIV virions, enhancing infection; this process is inhibited by EGCG, the peptide WW61, or surfen. Additionally, SEVI fibrils boost XMRV infection. Related references: PMID:18083097, PMID:19451623, PMID:19897482, PMID:21677644, PMID:19403677
  • Subcellularly, ACPP is found in secreted form, at the cell membrane, lysosome membrane, nucleus, cytoplasm, and cytosol.
  • Tissue expression is highest in prostate glandular and ductal epithelial cells, with additional expression in bladder, kidney, pancreas, lung, cervix, testis, ovary, and a weaker signal in a subset of other tissues. An alternative isoform is widely expressed, including in the sarcolemma of skeletal muscle.
  • Annotated with keywords including alternative splicing, amyloid, disulfide bond, glycoprotein, hydrolase, lipid metabolism, membrane, nucleus, and reference proteome.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to the N-terminal region (aa 1-100) of human ACPP. This design may enable recognition of both native and denatured protein, making it suitable for applications like WB and ELISA.
  • For Western blotting, consider testing whole cell lysates from human prostate cancer cell lines (e.g., LNCaP, PC-3) or transfected cells. The predicted molecular weight is approximately 45 kDa; however, glycosylation may result in a slight shift.
  • ELISA applications may benefit from using recombinant ACPP protein or peptide as capture antigen. The antibody’s cross-reactivity with mouse and rat allows for validation in rodent models, but confirm specificity in your experimental system.
