Biological Background: PCSK9 Function and Localization
- Proprotein convertase subtilisin/kexin type 9 (PCSK9), also known as neural apoptosis-regulated convertase 1 (NARC-1) or PC9, is a serine protease involved in cholesterol metabolism.
- Binds to low-density lipoprotein receptor family members (LDLR, VLDLR, LRP1/APOER, LRP8/APOER2), promoting their degradation in intracellular acidic compartments. Related references: PMID:18039658
- Enhances hepatic LDLR degradation via a clathrin/LDLRAP1-mediated pathway, preventing recycling or directing to lysosomes, and can induce LDLR ubiquitination. Related references: PMID:17461796, PMID:18197702, PMID:18799458
- Inhibits intracellular degradation of APOB and disposes of non-acetylated BACE1 intermediates in the early secretory pathway. Related references: PMID:18660751

Experimental Guidance and Technical Tips
- The immunogen corresponds to a peptide within amino acids 590-692 of human PCSK9, which includes part of the C-terminal domain. Consider that this region is present in the full-length proprotein and may be retained in some cleaved forms.
- For Western blotting (WB), the full-length PCSK9 has a predicted molecular weight of ~74 kDa. Additional bands corresponding to processed forms (e.g., mature ~62 kDa chain and prodomain) may be observed depending on sample preparation and cellular context.
- For immunoprecipitation (IP), validate the antibody’s performance with your specific sample type, as PCSK9 can be secreted or retained intracellularly.
- For immunofluorescence (IF/ICC) and immunohistochemistry (IHC-P), fixed and permeabilized samples are recommended due to the protein’s localization in multiple organelles. Validate in positive control tissues (e.g., liver, colon carcinoma) and include appropriate controls.
