  • Prolactin-inducible protein (PIP) is a secreted glycoprotein, also known as gross cystic disease fluid protein 15 (GCDFP-15), secretory actin-binding protein (SABP), and gp17.
  • The mature protein is a single-chain polypeptide of approximately 16.5–17 kDa, generated after cleavage of the signal peptide, and contains a pyrrolidone carboxylic acid at the N-terminus along with one intramolecular disulfide bond.
  • PIP possesses an actin-binding domain and is reported to interact with actin, suggesting a role in modulating the cytoskeleton or secretory pathways.
  • Expression is regulated by prolactin and androgens; it is found in pathological conditions of the mammary gland, including breast cysts and tumors, and is frequently used as an immunomarker for breast cancer.
  • In addition to the mammary gland, PIP is expressed in several normal exocrine tissues, such as the lacrimal, salivary, and sweat glands.
  • Functional studies and protein keywords indicate involvement in secretory processes and potential immune defense, though precise biological functions remain under investigation.

Experimental Guidance and Technical Tips

  • For immunofluorescence/immunocytochemistry (IF/ICC), consider using permeabilization if probing for intracellular PIP, but note that the protein is primarily secreted; surface staining on live cells may be possible without permeabilization.
  • In ELISA applications, this polyclonal antibody can be utilized as either capture or detection reagent; optimize coating and detection conditions for the specific assay format.
  • Validate cross-reactivity carefully in your sample system; this antibody is reported to react with human PIP. Cross-reactivity with other species has not been determined.
  • The observed molecular weight in Western blot may appear slightly larger than the predicted 16.5 kDa due to glycosylation.
