- Prokineticin-2 (PK2) is a secreted neuropeptide encoded by the PROK2 gene, also known as Bv8 homolog.
- It functions as a critical output molecule from the suprachiasmatic nucleus (SCN) that transmits behavioral circadian rhythm.
- Endogenous PK2 may also act locally within the SCN to synchronize neuronal output.
- PK2 potently contracts gastrointestinal (GI) smooth muscle, indicating a role in gut motility.
- The protein is secreted and contains a signal peptide and multiple disulfide bonds.
- Tissue expression is highest in the testis, with low levels detected in the small intestine.
- Alternative splicing produces isoforms, and mutations are associated with Kallmann syndrome and hypogonadotropic hypogonadism.
