Biological Background: Prohibitin Function and Localization
- Prohibitin 1 (PHB; PHB1) is a highly conserved protein with pleiotropic functions, including mitochondrial chaperone activity, nuclear transcription coregulation, and plasma membrane signaling, acting in a cell-compartment- and tissue-specific manner (PubMed:11302691, PubMed:20959514, PubMed:28017329). - In mitochondria, PHB forms large ring complexes with PHB2 in the inner membrane, functioning as a chaperone that stabilizes respiratory enzymes, regulates cardiolipin remodeling, and is essential for mitochondrial morphogenesis and longevity (PubMed:11302691).
