- Prohibitin (PHB1, PHB) is a pleiotropic protein with functions in mitochondria, nucleus, and plasma membrane, playing roles in cell signaling, mitochondrial integrity, and transcriptional regulation.
- In mitochondria, PHB forms large ring complexes with PHB2 in the inner mitochondrial membrane, acting as a chaperone that stabilizes respiratory enzymes and maintains mitochondrial morphogenesis, neuronal survival, and normal lifespan. The prohibitin complex regulates cardiolipin remodeling and OMA1 turnover, and functions as a mitophagy receptor targeting mitochondria for autophagic degradation.
