Biological Background: Prohibitin 2 Function and Localization
- Prohibitin-2 (PHB2), also known as BAP37, REA, or repressor of estrogen receptor activity, is a pleiotropic protein with cell-compartment- and tissue-specific functions.
- In mitochondria, PHB2 forms large ring complexes with PHB1 in the inner membrane, stabilizing respiratory enzymes and maintaining mitochondrial integrity. It regulates cytochrome-c oxidase assembly, acts as a mitophagy receptor, and mediates antiviral innate immunity by activating RIG‑I signaling. Related references: PMID:11302691 PMID:20959514 PMID:28017329 PMID:31522117
- In the nucleus, PHB2 functions as a transcriptional co-regulator, mediating repression by nuclear hormone receptors through histone deacetylase recruitment. It acts as an estrogen receptor-selective coregulator, potentiating antiestrogen effects and repressing estrogen activity.
- At the plasma membrane, PHB2 is involved in IGFBP6-induced cell migration and cooperates with CD86 in B lymphocytes to regulate IgG1 production. Upon CD40 engagement, it activates NF-kappa‑B signaling. Related references: PMID:24003225
- PHB2 is involved in human enterovirus 71 (EV‑71) infection by enhancing autophagy during the viral life cycle.
- Subcellularly, PHB2 localizes to the mitochondrial inner membrane, cytoplasm, nucleus, and cell membrane. It is expressed in myoblasts and undergoes phosphorylation and acetylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 1‑299 of human PHB2 (NP_001138303.1), covering the N‑terminal region. Verify sequence conservation with mouse and rat orthologs to support cross‑reactivity claims.
- For WB, the predicted molecular weight is ~33 kDa. Use appropriate positive controls, such as human cell lysates, and validate antibody specificity.
- For IHC‑P, consider optimizing antigen retrieval and antibody dilution using tissue known to express PHB2 (e.g., skeletal muscle).
- For IP, optimize antibody concentration and incubation conditions to ensure efficient capture of endogenous PHB2.
- For ELISA, recombinant protein or peptide corresponding to the immunogen can be used as coating antigen.
