Biological Background: Prohibitin 2 (PHB2) Function and Localization

  • Prohibitin-2 (PHB2), also known as Bap37, REA, and p22, is encoded by the PHB2 gene and is a member of the prohibitin family. It is a ubiquitously expressed protein with a molecular weight of ~33 kDa, found in multiple subcellular compartments including mitochondria, nucleus, cytoplasm, and plasma membrane.
  • In mitochondria, PHB2 forms large ring complexes with PHB1 in the inner mitochondrial membrane, serving as a chaperone that stabilizes respiratory enzymes and maintains mitochondrial integrity, essential for mitochondrial morphogenesis and neuronal survival. It also regulates cytochrome-c oxidase assembly (COX) and mitochondrial respiration. Related references: PMID:11302691 PMID:20959514
  • PHB2 functions as a mitophagy receptor, targeting damaged mitochondria for autophagic degradation, a process crucial for cellular quality control. Related references: PMID:28017329
  • In the nucleus, PHB2 acts as a transcriptional co-regulator, mediating repression by nuclear hormone receptors through histone deacetylase recruitment. It functions as an estrogen receptor (ER)-selective coregulator, potentiating antiestrogen activities and repressing estrogen activity.
  • At the plasma membrane, PHB2 is involved in IGFBP6-induced cell migration and cooperates with CD86 in B lymphocytes to regulate IgG1 production and NF-kappa-B signaling upon CD40 engagement. Related references: PMID:24003225
  • PHB2 is involved in host-virus interactions, enhancing the autophagy mechanism during human enterovirus 71 (EV-71) infection.
  • Post-translational modifications include acetylation and phosphorylation, and the protein undergoes alternative splicing, as noted in UniProt keywords.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 100-200 of human PHB2 (Q99623). This region may contribute to specific detection of PHB2 across human, mouse, and rat samples, but cross-reactivity should be validated in the relevant sample system.
  • For western blot (WB), expect a band at approximately 33 kDa. Use appropriate loading controls and validate antibody specificity with knockdown or knockout controls if possible.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimization of antigen retrieval and antibody dilution is recommended. Consider tissue-specific expression patterns, such as in myoblasts.
  • For immunoprecipitation (IP) and ELISA, titration of the antibody and use of suitable buffer conditions will be necessary. Ensure proper controls to confirm specific binding.
