Biological Background: Progranulin Function and Localization
- Progranulin (Granulin, GP88, PCDGF) is a secreted glycoprotein encoded by the GRN gene that acts as a key regulator of lysosomal function and as a growth factor involved in inflammation, wound healing, and cell proliferation. Related references: PMID:12526812 PMID:18378771 PMID:28073925
- It regulates protein trafficking to lysosomes and the activity of lysosomal enzymes, thereby facilitating the degradation of mature CTSD by CTSB. Related references: PMID:28453791 PMID:28541286
- As a wound-related growth factor, Progranulin directly promotes division, migration, and capillary-like tubule formation in dermal fibroblasts and endothelial cells.
- The protein blocks TNF-mediated neutrophil activation, preventing the release of oxidants and proteases that would otherwise damage epithelial cells. Related reference: PMID:12526812
- In the nervous system, Progranulin preserves neuronal survival, axonal outgrowth, and integrity, thereby modulating neuroinflammation. Related reference: PMID:18378771
- Subcellularly, Progranulin is found both secreted and within lysosomes, consistent with its dual role in extracellular signaling and intracellular degradation.
- Tissue expression is highest in kidney, with prominent presence in fibroblasts, epithelial cell lines, inflammatory cells, and bone marrow.
Experimental Guidance and Technical Tips
- The immunogen spans amino acids 20–310 of human Progranulin, covering the full-length precursor; it may recognize both the unprocessed protein and potentially processed granulin peptides. Consider validating selectivity in the relevant sample system.
- For western blotting, the predicted molecular weight of full-length Progranulin is ~64 kDa. Processed granulin peptides may appear as smaller bands; use appropriate positive and negative controls.
- In immunofluorescence/immunocytochemistry, note that Progranulin is primarily secreted and localized to lysosomes. Permeabilization and co-staining with lysosomal markers may enhance detection.
- Given the confirmed cross-reactivity with mouse, this antibody can be useful for studies in mouse models of inflammation, wound healing, or lysosomal storage disorders. Always validate in the intended species and application.
