Biological Background: Progastrin-releasing peptide Function and Localization
- Gastrin-releasing peptide (GRP) is a neuropeptide belonging to the bombesin family, encoded by the GRP gene. It is synthesized as a precursor (preproGRP) and processed into mature forms including GRP-10 and proGRP.
- GRP stimulates the release of gastrin and other gastrointestinal hormones, playing a key role in digestive system regulation.
- GRP is a potent mediator of itch (pruritus), contributing to nonhistaminergic itch sensation via mast cell degranulation and spinal cord itch signal transmission.
- In the central nervous system, GRP modulates fear memory consolidation in the amygdala and auditory cortex, and induces sighing via the pre-Bötzinger complex.
- GRP also regulates food intake and influences neuronal ion channels in the hippocampus, inhibiting voltage-gated sodium channels and enhancing potassium channels.
- Subcellularly, GRP is secreted and localized to cytoplasmic vesicles, secretory vesicle lumen, and neuron projections.
- Key post-translational modifications include amidation and cleavage on pairs of basic residues; alternative splicing produces multiple isoforms.
Experimental Guidance and Technical Tips
- This antibody was raised against recombinant human full-length progastrin-releasing peptide. For WB, a band around 16 kDa is expected; consider using reducing conditions and appropriate loading controls.
- For ELISA applications, optimize coating antigen, antibody dilution, and detection system based on your assay format.
- Cross-reactivity with mouse has been indicated, making it potentially useful in mouse model studies; however, validation in the relevant tissue or cell system is recommended.
