Biological Background: PRMT8 Function and Localization
- PRMT8 (Protein arginine N-methyltransferase 8) is a 45 kDa membrane-associated enzyme belonging to the protein arginine methyltransferase family, also designated HRMT1L3, HRMT1L4, and PRMT8.
- It is a brain-specific methyltransferase, with expression primarily restricted to the central nervous system.
- PRMT8 catalyzes the formation of monomethylarginine (MMA) and asymmetric dimethylarginine (aDMA) on substrates including histone H4, H2A, and H2A/H2B dimers, playing a role in chromatin remodeling and transcriptional regulation. Related references: PMID:16051612 PMID:17925405 PMID:26529540
- It also methylates EWS protein, although the biological outcome of this interaction is not fully understood. Related references: PMID:18320585
- The protein is anchored to the cell membrane via an N-terminal myristoylation motif, which is critical for its localization and function.
- Alternative splicing produces multiple isoforms, which may modulate its activity or substrate specificity.
- Structural studies have revealed a conserved methyltransferase domain and a unique N-terminal extension that distinguishes it from other family members.
Experimental Guidance and Technical Tips
- Given the immunogen encompasses amino acids 200-380 within the catalytic domain, the antibody is expected to recognize full-length PRMT8 and potentially some splice isoforms. Validate in your specific model system.
- For Western blot, consider using brain tissue lysates (human, mouse, or rat) as positive controls, since PRMT8 expression is brain-specific.
- In ELISA applications, as this is a polyclonal antibody, pairing with a PRMT8-specific monoclonal antibody may improve assay specificity if developing a sandwich ELISA.
