Biological Background: PRMT7 Function and Localization
- PRMT7 (Protein arginine N-methyltransferase 7) is a member of the protein arginine methyltransferase family, also designated as SBIDDS.
- Catalyzes both monomethylarginine (MMA) and symmetric dimethylarginine (sDMA) formation, exhibiting a preference for MMA generation.
- Symmetrically dimethylates histone H4 at Arg-3 (H4R3me2s), an epigenetic mark involved in chromatin regulation and gene imprinting.
- Methylates small nuclear ribonucleoproteins Sm D1 (SNRPD1) and Sm D3 (SNRPD3); this modification is essential for snRNP core particle assembly and biogenesis.
- In vitro, methylates histone H2A and myelin basic protein (MBP), although the in vivo significance remains to be fully established.
- Recruited by CTCFL at the H19 imprinted control region (ICR), where H4R3me2s may help recruit DNA methyltransferases.
- Localizes to both cytosol and nucleus, consistent with its roles in cytoplasmic and nuclear processes.
- Annotated with keywords including alternative splicing, chromatin regulator, methyltransferase, and disease variant (associated with dwarfism and intellectual disability).
Experimental Guidance and Technical Tips
- The immunogen maps to the C-terminal region (aa 513–692) of human PRMT7; the antibody may detect full-length PRMT7 and any isoforms or fragments containing this region.
- For Western blotting, consider reducing and denaturing sample conditions; the predicted molecular weight is 78 kDa, so use a molecular weight marker spanning the 70–100 kDa range.
- Cross-reactivity with mouse and rat PRMT7 has been reported; validate specificity and sensitivity in the relevant sample system before quantitative analyses.
- This antibody has been tested for WB and ELISA; other applications (e.g., IHC, IF) may require in‑house optimization.
- In ELISA, the recombinant protein immunogen can serve as a positive control to help establish a standard curve.
