Biological Background: PRMT6 Function and Localization
-
PRMT6 is a protein arginine N-methyltransferase that catalyzes asymmetric dimethylarginine (aDMA) formation, with a strong preference for aDMA over monomethylarginine (MMA). It targets arginine residues within glycine-arginine-rich motifs. Related references: PMID:17898714 PMID:18077460 PMID:18079182
-
PRMT6 specifically mediates asymmetric dimethylation of histone H3 at arginine 2 (H3R2me2a), a mark linked to transcriptional repression and mutually exclusive with activating H3K4 methylation. It also methylates histones H2A and H4 at arginine 3. Related references: PMID:17898714 PMID:18077460 PMID:18079182
-
PRMT6 acts as a transcriptional repressor of genes such as HOXA2, THBS1, and TP53, linking it to cellular senescence regulation. It also serves as a coactivator for steroid hormone receptors (ESR1, ESR2, PGR, NR3C1) and promotes gluconeogenic gene expression via CRTC2 methylation. Related references: PMID:19509293
-
PRMT6 regulates DNA base excision repair by methylating DNA polymerase beta (POLB), enhancing its DNA binding and processivity. Related references: PMID:16600869
-
It methylates non-histone substrates including HMGA1, HIV-1 proteins (Tat, Rev, NC), GPS2, and SIRT7, affecting processes such as chromatin architecture, antiviral defense, and mitochondrial biogenesis. Related references: PMID:16157300 PMID:16159886 PMID:17267505 PMID:30420520
-
PRMT6 is highly expressed in kidney and testis, and localizes predominantly to the nucleus.
-
PRMT6 is subject to phosphorylation and ubiquitin conjugation, and participates in alternative splicing regulation.
Experimental Guidance and Technical Tips
- The immunogen (recombinant protein covering amino acids 1-375) corresponds to the full-length PRMT6, which may generate a polyclonal response against multiple epitopes.
- In WB, a single band near 42 kDa is expected. Validate expression in cell lines or tissues known to express PRMT6, such as kidney or testis.
- For IF/ICC, nuclear localization should be observed. Include appropriate controls such as antigen-blocking peptides or PRMT6 knockout cell lines to confirm specificity.
- ELISA applications may be developed using the antibody as a capture or detection reagent; consider optimizing coating conditions and including a standard curve with recombinant PRMT6.
- Given the cross-reactivity with mouse, this antibody may be used to analyze PRMT6 in mouse samples; however, perform species-specific validation to ensure signal specificity.
