Biological Background: PRMT5 Function and Localization
- PRMT5 (Protein arginine N-methyltransferase 5), also known as JBP1, SKB1, and HRMT1L5, is a type II arginine methyltransferase that catalyzes both monomethylarginine (MMA) and symmetric dimethylarginine (sDMA) modifications (PubMed:10531356, PubMed:11152681, PubMed:11747828). Related references: PMID:10531356, PMID:11152681, PMID:11747828
- It methylates histones H2A, H3, and H4, regulating chromatin structure and gene expression, including repression at the PER1 promoter and gamma-globin loci (PubMed:25092918). Related references: PMID:25092918
- PRMT5 is critical for snRNP biogenesis by symmetrically dimethylating Sm proteins D1 and D3, necessary for spliceosome assembly and pre-mRNA splicing (PubMed:11747828, PubMed:12411503, PubMed:17709427). Related references: PMID:11747828, PMID:12411503, PMID:17709427
- Subcellularly, PRMT5 localizes to the cytoplasm, nucleus, chromosome, and Golgi apparatus, consistent with its diverse functional roles.
- It modulates signal transduction by methylating EGFR and RAF1, attenuating EGF-mediated MAPK signaling (PubMed:21258366, PubMed:21917714). Related references: PMID:21258366, PMID:21917714
- PRMT5 methylates p53/TP53, potentially influencing target gene specificity, and methylates various other substrates including MBD2, HOXA9, and Piwi proteins (PubMed:19011621, PubMed:22269951, PubMed:16428440). Related references: PMID:19011621, PMID:22269951, PMID:16428440
- Tissue expression is ubiquitous, and the protein undergoes acetylation and alternative splicing, with roles in biological rhythms and transcriptional repression.
Experimental Guidance and Technical Tips
- The immunogen spans residues 1-250 of human PRMT5; consider this region when evaluating potential isoform and species cross-reactivity.
- Applications include WB, IHC-P, IF/ICC, and ELISA; optimal working dilutions and conditions should be determined empirically for your specific experimental system.
- Cross-reactivity with mouse and rat makes this antibody a candidate for studies in rodent models; however, validation in the appropriate tissue or cell context is recommended.
- Given PRMT5’s ubiquitous expression, include appropriate controls (e.g., knockout or knockdown) to confirm signal specificity in your assays.
